Colony selection is an important step in
therapeutic protein discovery. Finding an optimized strain that can produce an
active and suitable biological product with high yield can significantly impact
large-scale production and downstream processes. We have developed a LC-MS/MS
based method that can rapidly and quantitatively screen colonies for producing
designed protein forms with high yield. This method can complement the
limitations of conventional colony selection methods such as western blot, ELISA
and
enzymatic assays, and offers significant advantage over those methods in the
case where immunological or enzymatic assays are not available or difficult to
carry out.
