Epitope mapping is an important tool in the discovery of high affinity and
high potency therapeutic antibodies. ProtTech provides a customized service for
epitope mapping.
Although the method of screening peptide library has been
used widely in epitope mapping, it has only achieved limited success. This is
because, in about 90% of cases, the antigenic determinant bound to an antibody
is not a linear stretch of amino acids but a uniquely folded tertiary structure,
which makes the epitope mapping a difficult task.
We have developed an epitope-mapping platform that can
protect the epitope regions of an antibody-antigen complex during limited
proteolytic digestion. We use a combination of specific and non-specific
proteolytic enzymes such as trypsin, Glu-c, pepsin, chymotrypsin to digest antibody-antigen complexes under
a well-controlled condition. The resulted peptides from each digest
are analyzed by LC-MS/MS, and the protection patterns are then analyzed by our
ProtTechEpiMap software to define the peptide regions that bound to antibodies.
